 【檢驗原理】

Diff 快速細胞染色液是採用世界衛生組織(WHO)推薦的染色方法而配製，它跟瑞氏染色(Wright Stain)一樣，都是利用Romanowsky Stain技術原理改良而成的，所得到的結果也跟瑞氏染色液相類似。但迪夫快速細胞染色所花費的時間較短，約1分鐘30秒即可完成染色步驟。較適合用於批量浸染，更加節省染色時間，且具背景清晰無沉渣等特點。

臨床應用與原理
耐酸染色液〈冷染法〉是一種用於肺結核桿菌
(Mycobacterium tuberculosis)/耐酸菌(Acid Fast Bacilli)染色之溶液。

傳統的耐酸染色液需要加熱使用，以促進染色效果，且
每隔一段時間，其中的複紅就必須過濾。而每一次過濾的
結果，都會降低複紅的染色效果。因此發現率普遍不高。
耐酸染色液係參考 Ziehl-Neelsen 法(a)配製而成，屬
於“冷染色法”，無須加熱，染色力強，使用簡單、方便，
是實驗室最佳的選擇。當複紅染色液滴到痰液塗片上，石
炭酸(Phenol)和鹽基性複紅(Fuchsin basic)所形成之複合物
會滲入到塗片上的所有微生物或細胞內部，使形成紅色。
加入酸性酒精進行脫色，除了肺結核桿菌/耐酸菌不會被脫
色外，其他的微生物或細胞皆會被脫色。脫色後，加入對
染液進行對染，除了肺結核桿菌/耐酸菌外，其他的微生物
或細胞皆會被染成藍色。
試劑成份
耐酸染色液〈冷染法〉包含三種成份：
1. 複紅(CARBOLFUCHSIN)：甲醇溶液中含Phenol (60
g/L)和Fuchsin Basic (25g/L) 。
2. 酸性酒精(ACID ALCOHOL)：含HCl(3%)和Methanol
(97%)。
3. 對染液(COUNTER STAIN)：甲醇溶液中含有Methylene
Blue(1.5 g/L)。

酸性酒精1X250ml
對染液1X250ml

保存及穩定性
1. 耐酸染色液〈冷染法〉可保存於 2-35℃。
2. 有效期內皆可使用。

檢體收集及準備
1. 測試檢體為病人之痰液。
2. 病人痰檢體可考慮使用痰前處理液(SPUTUM
PRETREATMENT SOLUTION)處理，以利染色之進
行及提高發現率。

注意事項
1. 本試劑為體外診斷試劑。
2. 所有痰檢體都須視為具有傳染性，須小心處理。
3. 超過效期，不能使用。

材料需求且提供
耐酸染色液〈冷染法〉。

材料需求但不提供
玻片，酒精燈和顯微鏡附油鏡。
操作方法
1. 將痰檢體製成塗片，略加熱固定〈塗片不可太厚，
否則會使脫色不完全，且塗片容易剝落〉。
2. 將複紅滴於塗片上，染色3-5 分鐘〈不須加熱〉。
3. 以自來水沖洗。
4. 將酸性酒精滴於塗片上，進行脫色，約1 分鐘。
5. 以自來水沖洗。若發現脫色不完全，可再以酸性酒
精脫色。
6. 將對染液滴於脫色後之塗片上，進行背景染色，約
10-30 秒。
7. 以自來水沖洗，風乾。
8. 置於顯微鏡下，以油鏡觀察。
結果判讀
紅色：肺結核桿菌/耐酸菌。
藍色：背景。
效能
適於肺結核桿菌/耐酸菌之染色用。

快速革蘭氏染色液 RAPID GRAM STAIN 試劑組合4X250ml/盒

臨床應用與原理
快速革蘭氏染色液是一種用於細菌染色之溶液。
細菌經過結晶紫及碘液染色後，根據是否可被脫色，可
分成二大類：脫色後，保有原來的顏色─藍色/黑色，即為
革蘭氏陽性菌，G(+)；脫色後，失去原來的顏色，且被沙
紅染成紅色，即為革蘭氏陰性菌，G(-)。
一般的革蘭氏染色液常常因為脫色時間控制不良，而造
成偽陰性出現，即革蘭氏陽性菌被染成革蘭氏陰性菌。而
且，染色所需的時間太長，約 4-5 分鐘。這些問題都是細
菌室長久以來所碰到的困難。快速革蘭氏染色液是由革蘭
氏染色法(Hucker modification)(a)修改而來。操作所需時間
極短，只要 40 秒即可完成所有步驟。準確性高，不易產
生偽陰性。

試劑成份 快速革蘭氏染色液含四種成份： 試劑組合4X250ml/盒
1. 結晶紫(CRYSTAL VIOLET)：含Crystal violet (7 g/L)。
2. 碘液(GRAM IODINE)：含I (35 g/L)和KI (70 g /L)。
3. 脫色液(DECOLORIZER)：含Isopropyl Alcohol (50%)和
Acetone (50%)。
4 沙紅(SAFRANIN)：含Sfranin (3 g/L)。

1. 快速革蘭氏染色液可保存於 2-35℃。

10. 吸乾或風乾，置於顯微鏡下，以油鏡觀察。
結果判讀
藍色/黑色：革蘭氏陽性菌，G(+)。
紅色：革蘭氏陰性菌，G(-)。

size:125ml 

Introduction:
Luxol-Fast blue staining of myelin/myelinated axons on formalin-fixed,
paraffin-embedded brain and spinal cord tissue sections, as well as frozen sections. The
myelin, including phospholipids, will be stained blue to green, and the neurons will be
stained violet. This stain is commonly used for identifying the basic neuronal structure
in brain or spinal cord tissue.

Kit Contents (for 125ml kit):

Luxol fast blue solution Ready to use, 125ml overnight 25-28℃
Cresyl violet solution Ready to use, 125ml 5-10 minutes. 25-28℃
lithium carbonate solution Ready to use, 125ml 30 seconds. 25-28℃
Control slide x 2 FFPE section -- 25-28℃
"Note:
 Preparing 5-7um thick sections will be more satisfactory."

Staining Protocol Recommendations:

1. Deparaffinize and hydrate to 95% ethyl alcohol (Frozen/vibratome sections may
need to have de-fat step: place sections directly into 1:1 alcohol/chloroform for
a few hours/overnight and then hydrate back 95% ethyl alcohol).
2. Leave in luxol fast blue solution in 56 C oven overnight (for frozen sections, not
longer than 16 hours).
3. Rinse off excess stain with 95% ethyl alcohol.

4. Rinse in distilled water.
5. Differentiate the slides in the lithium carbonate solution for 30 seconds.
6. Continue differentiation in the 70% ethyl alcohol for 30 seconds.
7. Rinse in distilled water.
8. Check microscopically to see if gray matter is clear and white matter sharply
defined.
9. Repeat the differentiation steps (step 5-7) if necessary.
10. When differentiation is complete, place in distilled water.
11. Counterstain in the cresyl violet solution for 5-10 minutes.
12. Rinse in distilled water (don’t rinse in 70% alcohol which will clear off fast blue
staining).
13. Differentiate the slides in 95% ethyl alcohol for 5 minutes (check
microscopically).
14. 100% alcohol 2x5 min.
15. Xylene 2x5 min.
16. Mount with resinous medium.
Results:
Myelin, including phospholipids ------------------ blue to green
Neuron ---------------------------------------------- pink to violet
